Influence of ferrocene on trypsin activity

Abstract

Many transition metals and their complexes have shown inhibitory effect on some proteolytic enzymes, including trypsin. Their inhibitory activity is based on the direct binding to the active site of trypsin, mimicking formation of a five-coordinate transition state of the metal ion required for reaction. Ferrocene was selected as a potential inhibitor because it belongs to the family of organometallic, so called "sandwich" compounds, and its cyclopentadienyl rings might have an adequate geometry. The influence of ferrocene on trypsin activity using BAPNA (N-α-benzoyl-DL-arginine p-nitroanilide) as a substrate was investigated, as well as the influence of ferrocene on growth of Esherichia coli ATCC 25922. It was found that ferrocene decreases specific activity of trypsin and also has an inhibitory affect on growth of Esherichia coli.
