Ćelijska signalna transdukcija - modulacija slobodnim radikalima

Sažetak

Signalna transdukcija je proces prenosa signala sa ćelijske površine u unutrašnjost ćelije. Ovaj ubikvitarni mehanizam omogućava obavljanje osnovnih ali i ćelijsko-tkivno specifičnih procesa koji učestvuju u održavanju ćelijske homeostaze. Narušavanje normalnih transdukcionih procesa je najčešća molekulska baza u patogenezi i razvoju komplikacija mnogih bolesti. Takođe, poznavanje svih činioca uključenih u kaskade signalnih puteva otvara mogućnost primene različitih modulatora u terapijske svrhe. Jedan od najbolje proučenih puteva signalne transdukcije je aktivacija fagocitne NADPH oksidaze. Međutim reaktivne kiseonične vrste koje nastaju u ovom procesu mogu modulisati brojne ćelijske transdukcione puteve. Šećerna bolest je praćena intenziviranim oksidativnim stresom koji dovodi do trajnog oštećenja tkiva. Oksidaciono oštećenje može da pokrene različite signalne kaskade koje modulišu ćelijske funkcije i mogu dovesti do ćelijske smrti. Hiperglikemijom indukovana aktivacija PKC-MAP kinaznog puta danas se smatra ključnim biohemijskim događajem u razvoju dijabetesnih komplikacija i insulinske rezistencije.
