Fosforilacija c-Jun-N-terminalnih kinaza i nuklearni transport Hsp70 proteina u hipokampusu Wistar pacova izloženih stresu

Sažetak

Glukokortikoidi su ključni regulatori odgovora hipokampusa na neuroendokrini stres. Njihovo delovanje je povezano sa aktivacijom podfamilije mitogen-aktiviranih kinaza (MAPK), nazvanom c-Jun-N-terminalne kinaze (JNK) čija aktivacija koreliše sa pojavom neurodegeneracije. Odgovor na stres takodje uključuje aktivaciju ćelijskih protektivnih mehanizama preko članova proteina toplotnog šoka (HSP) koji su medijatori neuroprotekcije. U ovom radu pratili smo signale JNK i Hsp70 u citoplazmi i jedru hipokampusa Wistar pacova koji su izlagani akutnom, hroničnom ili kombinovanom stresu. Aktivnost JNK1 smanjivala se u oba ćelijska kompartmana pod delovanjem sva tri stresa, ali je aktivnost JNK2/3 u citoplazmi rasla u akutnom stresu i bila je ili nepromenjena ili snižena u hroničnom i kombinovanom stresu. Istovremeno u uslovima sva tri stresa zapažena je značajno povišena translokacija Hsp70 u jedro. Naši rezultati ukazuju na mogućnost da je proneurodegenerativna signalizacija JNK u navedenim stresnim uslovima uravnotežena prisustvom povećane količine Hsp70 u jedru ćelija hipokampusa, što je naročito ispoljeno u hroničnom stresu.
