Sažetak
Lycoperdon perlatum (Pers) 1796 je saprobna puhara sa širokom rasprostranjenošću. Jestiva je kao mlada, kada je gleba još celovita i bela. Zbog prijatne teksture i ukusa, koristi se u supama, kao zamena za knedle. Cilj ovog rada bio je da se prouči bioaktivnost sirovih ekstrakata vrste L. perlatum pripremljenih od samoniklih sporokarpa poreklom iz istočne Srbije (Sikole, selo blizu Negotina) tokom 2012. godine. Skrining bioaktivnosti obuhvatio je antioksidativnu (testovi DPPH. i FRAP), antiproliferativnu (ćelijska linija humanog adenokarcinoma dojke, MCF-7; testovi MTT i SRB) i antibakterijsku aktivnost (tri referentna ATSS soja; mikrodilucioni test). Polarniji ekstrakti ispitivane gljive (vodeni - LycAq i etanolni - LycEtOH) testirani su na antioksidativnom i antitumorskom nivou, dok je nepolaran ekstrakt (LycHex) testiran na antibakterijskom nivou. Osim toga, LycAq i LycEtOH hemijski su preliminarno okarakterisani (sadržaj ukupnih fenola i flavonoida) pomoću UV-VIS spektrofotometrije. Najveća anti-DPPH radikalska aktivnost uočena je za LycAq (IC50 = 46.56 µg/ml). U poređenju sa LycAq, manje polarni ekstrakt LycEtOH pokazao je neznatno bolji potencijal za redukciju gvožđa (IC50 = 21.87 µg/ml i IC50 = 19.28 µg/ml). Međutim, sadržaj ukupnih fenola u oba ekstrakta bio je sličan (≈ 2.0 mg GAE/g.d.w.). S druge strane, krajnje blaga antibakterijska aktivnost uočena je na soj Staphylococcus aureus ATCC 25922 (LycHex, MIC = 3.12 mg/ml) i ćelije MCF-7 (uz najznačajniju aktivnost za LycEtOH nakon 72 h, IC50 = 367.54 µg/ml i IC50 = 390.03 µg/ml, na testovima MTT i SRB). Prema dobijenim eksperimentalnim podacima, L. perlatum može se smatrati dobrim izvorom novih i potentnih prirodnih antioksidanasa sa potencijalnom primenom u redovnoj ishrani.
