Molecular characterization of turnip yellows virus isolates from canola in Serbia

Sažetak
Tokom novembra 2019. godine, simptomi koji upućuju na prisustvo virusa žutice postrne repe (turnip yellows virus, TuYV) primećeni su u mnogim usevima uljane repice gajenih širom Srbije. U cilju identifikacije prouzrokovača oboljenja, sakupljeno je ukupno 206 uzoraka, koji su serološki testirani na prisustvo TuYV, virusa mozaika karfiola (cauliflower mosaic virus, CaMV) i virusa mozaika postrne repe (turnip mosaic virus, TuMV) korišćenjem komercijalnih DAS-ELISA kitova. Prisustvo TuYV serološki je dokazano u 91,75% testiranih uzoraka sakupljenih na 24 lokaliteta gajenja u svih sedam pregledanih okruga, dok prisustvo TuMV i CaMV u testiranim izorcima uljane repice nije potvrđeno. Dalja molekularna karakterizacija sedam odabranih ELISA pozitivnih uzoraka na osnovu delimične sekvence P0 gena ukazala je na visok stepen nukleotidne identičnosti izolata TuYV prikupljenih tokom 2019. godine, kao i na njihovu veliku sličnost sa prethodno identifikovanim izolatima ovog virusa poreklom iz kupusa i slačice iz Srbije. Filogenetska analiza je pokazala grupisanje izolata TuYV iz Srbije u TuYV/BrYV grupu. Međutim, nukleotidna i aminokiselinska sličnost sekvenci svih izolata iz Srbije, kao i filogenetska analiza, ukazuju na postojanje dve populacije ovog virusa u našoj zemlji. Dalja istraživanja treba usmeriti ka utvrđivanju varijabilnosti populacije TuYV u Srbiji analizom sekvenci celog genoma što će doprineti boljem razumevanju epidemiologije ovog patogena, a sve u cilju razvoja i primene adekvatnih mera kontrole.
